What are midi-chlorians?
Without the midi-chlorians, life could not exist, and we would have no knowledge of the Force. They continually speak to us, telling us the will of the Force. When you learn to quiet your mind, you'll hear them speaking to you.

What is midi-chlorian manipulation?
Plagueis was finally learning to do that, and was just beginning to learn how to persuade, prompt, cajole, and coax them into action. Already he could command them to promote healing

the host's midi-chlorians as well. Your dark presence will corrupt them.
Then we have the matter of Dark Transfer. Krayt uses it on Cade and kills him, then brings him back to life.
And then we have Muur's rakghouls. Exar Kun's terentateks.
All these were done by influencing midi-chlorians and manipulating them.
In fact the book admits the fact that ancient Sith could have been capable of doing what Plagueis has done if not for their fumbling ignorance of the Cosmic Force.
In the same way that the pre-Bane Sith had been responsible for their own extinction, the great dark side Lords of the past had doomed themselves to the nether realm through their attempts to conquer death by feeding off the energies of others, rather than by tapping the deepest strata of the Force and learning to speak the language of the midi-chlorians.
